Can I refer this way only to Anna (respectfully) or the sentence definitely refers to multiple female persons?

Hi Bunew,

In written form, if you addressed just one person it would be -

Vous êtes vendeuse? ( without the -s)

But in spoken French, there's no way to distinguish between the two as they sound the same but the context would allow you to be understood.
